Is it possible to make ashop and when someone click buy he get the item and his points instantly decreased

I don't think so, I think you have to manually take away points. On my forum, I (or an admin) will have to go into the admin panel and manually decrease the number of points.

I use EmailMeForm for the shop, and then I just deduct their points after completing their purchase.

You can re-create an actual shop though with EmailMeForm - it also sends their IP with the form so you can manually make sure that whoever bought the item is actually the user and not someone else, also it can be used to block them from your website, etc.
